Well, for her now and me 2 years ago (well okay, actually there was no pronoun field on the Shard 2 years ago, there was a gender field instead but same general idea), it was one of the first steps we took after we realized we were trans. I don't want to put words in her mouth, but for me, it was me saying publicly for the first time "I am a girl," even if it was only on this little forum. 

Although for me, while changing that was exciting, getting actually referred to with feminine pronouns is what really made me happy, but again, I'm just one trans girl among millions, I can't speak for all of us.

did it before I became trans. Felt more comfortable. More in line with who I wanted to be.
