If someone who had the potential to become a Misting or Mistborn was given a Hemalurgic spike before they had the opportunity to snap and come into their powers, would the Connection to Ruin prevent them from snapping and gaining their inborn Allomantic powers from Preservation's power?

Era 2? They'd snap from the trauma of having a spike stuck in them as a child, and I doubt it would matter anyways. Era 1? I don't know. Maybe, but Preservation and Ruin aren't opposites, and connection to one automatically prevent connection to the other.
