I have a couple of questions. Excuse me, if already answered.

1. To create a Kandra or a Koloss, we need a metallic Spike that is already charged, right? If so, do we know, how are the Kandras' “Blessings” charged?

2. It is said that Kandras are “of Preservation”, whereas the Kolosses are “of Ruin” — I don't remember how exactly it was put. Was it only figurative, or is there any Shard-a-matic analogy?

3. Is it possible that the most important difference (or one of them) between the two creature types is that Kolosses' Spikes where charged by killing a person with the specific trait and Kandras' Spikes were charged with another, less “invasive” way (e.g. just by Cosmeric kind of Goddidit)?

People are murdered with them and their spark of life goes into the spike. You can steal more than just Allomancy/Feruchemy with Hemalurgy. You can in fact steal anything related to the soul, apparently.

Kandra and Koloss are both born of Hemalurgy, but Shard-a-matically (Realmatically), I believe neither are really of Ruin, though it's possible that the spikes stole Ruin's Investiture specifically from people. The reason they're 'of Ruin' is because Ruin could control both thanks to the use of Hemalurgy (his magic system). Everything sentient on Scadrial pre-Sazed had an imbalance of Preservation, as far as I know. After Sazed, though, there's some very convincing speculation that the koloss-blooded have an imbalance of Ruin rather than Preservation to make them sentient.

Nope. Kandra spikes came from killing people. I think it's possible for Shards to directly create Hemalurgic spikes that are pre-filled though. I suspect this is what TLR did to originally create the kandra, but I am less than confident of this assertion, since I am not sure if he just turned the Feruchemists into mistwraiths and spiked them later, or spiked them during his use of the Well. In any case, the books directly say that later kandra were created by killing people (I think?).

Pretty sure the HoA states that the 1st Generation was changed to Mistwraiths and then spiked by TLR. 

 

As a reminder, someone who has something stolen doesn't have to be killed to have their power/whatever stolen, per WoB, though that is usually the case because of who is doing it and the strike point of the spike.

The Kandra say they are of Preservation, not because they use Preservation's magic, but because they hide Ruin's body. Plus, they are all trained to remove their spikes at critical junctures.
