If a Shardblade were to cut an Inquisitor, would it kill them? I mean, Inquisitors don't have souls and Shardblades cut through people's souls so I'm not sure on this one.

Inquisitors have souls.  Twisted and heavily altered souls, but souls nonetheless.  Therefore, a Shardblade injury of sufficient severity would indeed kill an Inquisitor.  (It'd skip the pyrotechnics, though)

The only thing that would make an Inquisitor Shardblade-proof would be if they have F-Gold and a large health reserve, because we know Bloodmakers can theoretically heal Shardblade wounds. Under normal circumstances however, they'd be just as dead from Shardblade-to-spine as anyone else.

Oh, and setting aside that Inquisitors aren't soulless and so that's a nonstarter, even if they were it wouldn't make them Shardblade-proof; if they're not considered 'alive' then the Shardblade would just cut them on the Physical plane the same it does anything considered 'dead'.

See also: Twisted and heavily altered.  Inquisitors' souls have essentially been drawn into upper and lower halves, with a Linchpin spike connecting the two. Severing the connection, either by Shardblade cut or by spike removal, would kill the Inquisitor.
